การสร้างเอนไซม์ของเชื้อราสาเหตุโรคทางใบของไม้ประดับ

บทคัดย่อ

การศึกษาครั้งนี้มีวัตถุประสงค์เพื่อตรวจสอบความสามารถของเชื้อราสาเหตุโรคทางใบของไม้ประดับใน
การสรา้ งเอนไซม์ย่อยสลายผนังเซลล์พืช (cellulase, pectinase) และเชื้อรา (chitinase) บนอาหารแข็ง ในสภาพ
ห้องปฏิบัติการ โดยแยกเชื้อราจากใบพืชที่แสดงอาการโรค ทั้งหมด 13 วงศ์พืช (32 ไอโซเลท) ภายในสถาบันเทคโนโลยี
พระจอมเกล้าเจ้าคุณทหารลาดกระบัง กรุงเทพฯ และทำการทดสอบกิจกรรมการสร้างเอนไซม์ cellulase และ
pectinase ประเมินโดย hydrolysis capacity (HC) value ซึ่งได้จากสัดส่วนของ hydrolysis zone และเส้นผ่าน
ศูนย์กลางโคโลนี ในขณะที่ chitinase ประเมินจากขนาดเส้นผ่านศูนย์กลางโซนสีม่วงบนอาหาร colloidal chitin
จากผลการทดลอง พบว่าเชื้อราทดสอบทุกไอโซเลท (ยกเว้น Co 211-1) สามารถสร้าง cellulase ได้ในระดับต่ำ ซึ่งมี
ค่า HC value น้อยกว่า 1.00 สำหรับ pectinase พบการสร้างเอนไซม์ได้ในระดับต่ำ มีค่า HC value น้อยกว่า 1.00
ในทุกไอโซเลท ยกเว้น กลุ่มของเชื้อรา Colletotrichum spp. 19 ไอโซเลท, Bipolaris sp. (Bi 21-2) และ Nigrospora
sp. (Ni 12-4) ที่ไม่พบการสร้างเอนไซม์ดังกล่าว และการทดสอบ chitinase พบว่าเชื้อรา Colletotrichum spp.
ทุกไอโซเลท (ยกเว้น Co 211-1) และ Alternaria spp. จำนวน 2 ไอโซเลท สามารถสร้างเอนไซม์ได้ระดับปานกลาง
ขณะที่เชื้อรา Cuvularia spp. จำนวน 4 ไอโซเลท และ Bipolaris sp. (Bi 21-2) พบกิจกรรมการสร้างเอนไซม์ในระดับ
ต่ำ และไอโซเลทอื่นๆ ไม่พบกิจกรรมการสร้างเอนไซม์ chitinase จากนั้นทำการทดสอบความสามารถในการทำให้เกิด
โรคโดยวิธี detached-leaf test ผลการทดลองที่กล่าวมาข้างต้น พอสรุปได้ว่า เชื้อราทดสอบ (จากพืชอาศัยที่ต่างกัน)
ภายในสกุลเดียวกันมีศักยภาพในการสร้างเอนไซม์ที่ย่อยสลายผนังเซลล์พืชไม่แตกต่างกัน
