บทคัดย่อ
ความแปรปรวนทางพันธุกรรมเป็นข้อมูลสำคัญที่จะเป็นประโยชน์ต่อการดำเนินงานการอนุรักษ์และฟื้นฟูประชากรปลาท้องถิ่นในแหล่งนํ้าธรรมชาติ การศึกษานี้จึงประเมินความแปรปรวนทางพันธุกรรมของ
กลุ่มตัวอย่างพ่อพันธุ์และแม่พันธุ์ปลาเลียหินที่นำมาเพาะพันธุ์เพื่อปล่อยลูกพันธุ์สู่แหล่งนํ้าธรรมชาติบริเวณลำนํ้ากอน (ลำนํ้าสาขาของแม่นํ้าน่านตอนบน) โดยเครื่องหมายไมโครแซทเทลไลท์ 5 ตำแหน่ง
ผลการศึกษาพบว่าเครื่องหมายทั้ง 5 ตำแหน่ง มีความหลากรูปแบบ (Polymorphic) โดยพ่อพันธุ์ และแม่พันธุ์ มีจำนวนอัลลีลต่อตำแหน่ง (A) 10.4 และ 10.2 ค่า Allelic richness (Ar) 10.31 และ 9.96
ค่าเฮตเทอโรไซโกซิตี้จากการสังเกต (Ho) 0.58 และ 0.67 และค่าเฮตเทอโรไซโกซิตี้จากการคาดหมาย (He) 0.82 และ 0.83 ผลรวมที่เครื่องหมายทุกตำแหน่งกลุ่มตัวอย่างพ่อพันธุ์และแม่พันธุ์อยู่ในสมดุล
ฮาร์ดี - ไวน์เบิร์ก สำหรับผลการวิเคราะห์ความต่างทางพันธุกรรมพ่อ พันธุ์และแม่พันธุ์มีค่าระยะห่างพันธุกรรม 0.512 และมีค่าสัมประสิทธิ์เอฟ FST 0.0621 (95 % CI 0.039 - 0.091) ผลการประเมินจำนวน Effective Population Size (Ne) ทั้ง 2 กลุม่ ตัวอย่างมี Ne ไม่จำกัด ผลจากการศึกษานี้แสดงถึงประชากรปลาเลียหินในลำนํ้ากอนมีความเหมาะสมต่อการนำมาเพาะพันธุ์เพื่อการฟื้นฟูประชากรธรรมชาติในพื้นที่ศึกษา


Abstract
The genetic variation is the important data for restoration and conservation of local fish population. This study evaluated the genetic variation of Garra cambodgiensis broodstocks in Kon River, the tributary of upper Nan River. The finding showed that all of the five microsatellite loci were polymorphic. The genetic variation within male and female populations were not different, the average number of allele per locus (A) 10.4, 10.2, allelic richness (Ar) 10.31, 9.96, observed heterozygosity (Ho) 0.58, 0.67 and expected heterozygosity (He) 0.82, 0.83 for male and female respectively. At all loci both populations were in Hardy-Weinberg equilibrium. The genetic differentiation between male and female, the genetic distance = 0.512 and FST  = 0.0621 respectively (95 % CI 0.039 - 0.091). Moreover, the effective population size (Ne) in both samples were infinite. Based on the results of this study, the broodstock are suitable for breeding plan and population restoration in Kon River.
